To say exactly where a known thing is, you put está (or están) and then a preposition of place. The general one is en: in, on and at all at once – el libro está en la mesa, Ana está en casa. The precise ones are mostly two words plus de: al lado de la puerta, detrás de la casa, cerca de la plaza.

Two contractions: al and del
When a or de meets the article el, the two words always fuse: a + el = al, de + el = del. There is no choice about it – a el and de el are simply not written. It happens only with el: a la, de la, a los, de las stay as two words. Cerca del banco, but cerca de la farmacia; vamos al cine, but vamos a la playa.

Right and left
A la derecha is on the right, a la izquierda on the left; add de to say right or left of something: el baño está a la derecha, la cocina está a la izquierda de la entrada. Entre joins its two points with y: entre la farmacia y el banco. And enfrente de (opposite, across the street) is a close cousin of delante de that you will hear in every set of directions.
El supermercado está al lado del banco, a la derecha.The supermarket is next to the bank, on the right.
Las llaves están encima de la mesa, entre el teléfono y el café.The keys are on the table, between the phone and the coffee.
